Is this occurring with Tx STMS or are you using OS commands?

Dear All,
when we are transporting request getting error "Errors: connect failed due
to DbSL load lib failure"
I have checked ittollbox and got if i will run "NTENV2REG.EXE" then problem
may be resolve.
Pls suggest to run this downtime is required or i can run even if SAP is up.
Pls guide clearly or tell other solution
