[sap-basis] Transport control program tp ended with error code 0249

Dear All,
when we are transporting request getting error "Errors: connect failed due to DbSL load lib failure"
I have checked ittollbox and got if i will run "NTENV2REG.EXE" then problem may be resolve.
Pls suggest to run this downtime is required or i can run even if SAP is up.
Pls guide clearly or tell other solution
