Are you posting a customer transaction or are you posting a clearing
transaction ?
If it is a customer invoice or a credit memo, you will be forced to enter a
profit center in at least one of the line items. If it is a clearing
transaction, you have no choice to enter a profit center at all in any line
item.
In any case, please do an FB03, view the document ( after tweaking the lay
out so as to include the profit center field ) via the G L view and see
what happens.

In document splitting you applied inheritance rule (in which it will take
profit center from previous transaction also such as you raise an bill for
a customer and in that bill you gave profit center now you are receiving
payment against that bill so in that case system will automatically take
profit center which is mentioned in your bill)
